The effect of Cherry laurel (Laurocerasus officinalis Roem.) leaf extract on the growth, survival and some antioxidant enzymes of rainbow trout (Oncorhynchus mykiss Walbaum, 1792) exposed to oxidative stress induced by high temperature

Öz

In this study, the effects of cherry laurel (Laurocerasus officinalis) leaf extract on the growth parameters, survival rate and some antioxidant enzymes [superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx)] of the rainbow trout (Oncorhynchus mykiss) in oxidative stress induced by high temperature were investigated. For this purpose, two trials, one of which was a preliminary trial, were conducted. In the preliminary study, the effect of cherry leaf extract supplemented to the diet with different concentrations (1, 5, 10, 15 g extract/kg) on growth, survival rate and antioxidant enzymes in blood and gill tissue were examined and the optimum concentration was determined as 15 g/kg. In the other study, the effect of supplementation of cherry laurel leaf extract with a concentration of 15 g/kg food to the diet on growth, survival rate, feed consumption, biochemical composition and antioxidant enzymes in the liver were investigated in juvenile trout induced by three temperature applications of 15, 19 and 21°C. The used cherry laurel leaf extract caused growth retardation in 15oC but showed a positive effect on growth in 21 oC. Also, survival rate increased significantly in the groups fed with cherry laurel supplemented diets in 19 and 21oC (p<0.05). In terms of antioxidant enzymes, there were no significant difference between the groups (p>0.05). Considering the results obtained at the end of the study, it was determined that cherry laurel increased the survival rate of trout under stress. As a result, it can be said that cherry laurel which is economic and easily found plant have a potency to become a new antioxidant substance that prevent losses by adding to the trout diet during the summer heat increase in aquaculture production.

Öz

Bu çalışmada, yüksek sıcaklıkla oksidatif strese maruz bırakılan gökkuşağı alabalığında (Oncorhynchus mykiss), karayemiş yaprağı (Laurocerasus officinalis) ekstraktının büyüme parametreleri, yaşama oranı ve bazı antioksidan enzimler [süperoksit dismutaz (SOD), katalaz (CAT) ve glutatyon peroksidaz (GPx)] üzerine etkisi incelenmiştir. Bu amaçla, biri ön deneme olmak üzere iki çalışma yürütülmüştür. Ön denemede yeme çeşitli konsantrasyonlarda (1, 5, 10 ve 15 g ekstrakt/kg) katılan karayemiş yaprağı ekstraktının büyüme, yaşama oranı, kanda ve solungaçta antioksidan enzimler üzerine etkileri incelenerek, uygun karayemiş yaprağı ekstraktı konsantrasyonu 15 g/kg yem olarak belirlenmiştir. Diğer denemede ise 15, 19 ve 21oC olmak üzere 3 sıcaklık uygulaması ile oksidatif stres oluşturulan jüvenil gökkuşağı alabalığında 15 g/kg konsantrasyonunda yeme ilave edilen karayemiş yaprağı ekstraktının büyüme, yaşama oranı, yem tüketimi, biyokimyasal kompozisyon ve karaciğerde antioksidan enzimler üzerine etkileri incelenmiştir. Kullanılan karayemiş ekstraktının büyümeyi 15 oC’de gerilettiği, 21 oC’de ise olumlu yönde etkilediği tespit edilmiştir. Ayrıca 19 ve 21°C’de karayemiş ilaveli yemle beslenen gruplarda, yaşama oranının kontrol gruplarına göre önemli oranda arttığı belirlenmiştir (p<0.05). Enzim analizleri bakımından her iki çalışmadaki gruplar arasında istatistiksel olarak önemli bir fark bulunmamıştır (p>0.05). Çalışmanın sonunda elde edilen sonuçlar göz önünde bulundurulduğunda, karayemişin stres altında yaşama oranını arttırdığı belirlenmiştir. Sonuç olarak ekonomik ve kolay bulunan bir bitki olan karayemişin, su ürünleri yetiştiriciliğinde yaz aylarındaki sıcaklık artışı süresince yeme ilave edilerek yaşanan kayıpları engelleyen yeni bir antioksidan madde olma potansiyeline sahip olduğu söylenebilir.
