Haematococcus pluvialis Flotow (Chlorophyceae)'un Farklı Işık Şiddetlerinde Vejetatif Büyüme Özellikleri.

Öz

Haematococcus pluvialis’in vejetatif kültürlerinde, optimal ışık şiddeti aralığı ve büyüme performanslarının tespiti için 50, 100, 200, 400 ve 600 µmol foton m-2 sn-1’lik beş farklı ışık şiddeti uygulandı. Deneme gruplarında kuru ağırlık ve toplam karoten miktarları artan ışık şiddetleri ile doğru orantılı olarak artarken, toplam klorofil miktarı 200 µmol foton m-2 sn-1’lik aydınlatmaya kadar arttı (2.21 mg L-1)ve bunun üzerindeki aydınlatma şiddetlerinde herhangi bir değişim görülmedi (ortalama 2.35 mg L-1). Hücre sayısı tüm gruplarda karanlık periyoda denk gelen 15. saatte ortalama 3.45 x 104 hücre ml-1’den 18. saatte 5.66 x 104 hücre ml-1’ye arttı. Sonuç olarak, 200 µmol foton m-2 sn-1’lik ışık şiddetinin üzerindeki aydınlatmalarda astaksantin birikiminin tetiklendiği ve hücre sayısının azaldığı görüldü (50, 100, 200, 400 ve 600 µmol foton m-2 sn-1’lik ışık şiddetleri için sırasıyla 12.73 x 104, 16.10 x 104, 19.73 x 104, 15.90 x 104 ve 12,89 x 104 hücre ml-1). Beş farklı ışık şiddetinin uygulandığı denemede hücrelerin vejetatif safhada kültür edilebilmesi için optimal ışık şiddeti aralığı 50 - 200 µmol foton m-2 sn-1 olarak bulundu ve en iyi büyüme 200 µmol foton m-2sn-1’lik ışık şiddetinde gerçekleşti
