Determination of Phytoplankton Pigment Concentration and Size Classes Using HPLC Technique (South-Eastern Black Sea-Rize)

Abstract

In the present study, seasonal variations of phytoplankton and pigment composition from May 2014 to April 2015 were investigated along the south-eastern coasts (Rize) of the Black Sea. Surface (0,5 m) water samples were collected from five different stations (Iyidere, Rize, Cayeli, Pazar and Findikli station) during the study period. A total of 71species were identified during the study period, 41 species of these were dinoflagellate species, 18 species were diatoms and 12 species were other groups (silicoflagellate, euglenophyta, prymnesiophyta, chlorophyta etc.). In terms of species numbers, dinoflagellates contributed to 57,76% of the total phytoplankton species. Diatoms were the second important group with the contribution of 25,35% to the total phytoplankton species, and followed other groups with 16,89%. The dominant pigment in the area was chlorophyll-a (mean 1,57 μg/L) revealed a consistency with phytoplankton cell numbers. The second prominent pigment was fucoxanthin (mean 0,35 μg/L) and followed by 19-Hexanoyloxifucxanthin (mean 0,24 μg/L) and peridinin (mean 0,21 μg/L). The positive relationships between cell number and pigment concentrations revealed that these marker pigments can be used for taxonomic characterisation of phytoplankton groups.

Fitoplankton Pigment Konsantrasyonunun ve Boy Gruplarının HPLC Tekniği ile Belirlenmesi (Güneydoğu Karadeniz, Rize)

Abstract

Mayıs 2014-Nisan 2015 tarihleri arasında Güney Doğu Karadeniz kıyılarında (Rize) yürütülen bu çalışmada, fitoplankton ve pigment konsantrasyonunun mevsimsel değişimleri araştırılmıştır. Araştırma süresince beş farklı istasyonda (İyidere, Rize, Çayeli, Pazar ve Fındıklı istasyonu) yüzey suyundan (0,5 m) örneklemeler yapılmıştır. Çalışmada, dinoflagellatlar tür sayısı bakımdan en fazla gözlenen fitoplankton grubu olmuştur. Çalışma dönemi boyunca toplam 71 tür belirlenmiş, bunların 41’i dinoflagellat, 18’i diatom ve 12’si de diğer gruplar (silikoflagellata, öglenofita, primnesiofita, klorofita vb.) altında değerlendirilmiştir. Kalitatif olarak toplam fitoplankton tür sayısında grupların yaptığı katkı değerlendirildiğine dinoflagellat %57,76 ile ilk sırada yer alırken, bunu %25,35 ile diatom türleri ve %16,89 ile diğer gruplara ait türler oluşturmuşlardır. Bölgede baskın pigment klorofil-a olup (ortalama 1,57 μg/L) fitoplankton hücre sayısı ile uyum içerisinde olduğu tespit edilmiştir. Klorofil-a’dan sonra en baskın pigment fukoksantin olup (ortalama 0,35 μg/L) bunu sırasıyla 19’-Heksanoloksifukoksantin (ortalama 0,24 μg/L) ve peridinin pigmenti (ortalama 0,21 μg/L) takip etmiştir. HPLC sonuçlarına göre pigment konsantrasyonları ile hücre sayıları arasında tespit edilen pozitif ilişkiler marker pigmentlerin fitoplankton gruplarını taksonomik açıdan karakterize etmek için kullanılabileceğini göstermiştir.
